چکیده

Hyaluronate (HA) distribution patterns were examined in the cranial mesenchyme underlying the mesencephalic neural folds of mouse embryos maintained in roller tube culture. Using standard image-processing techniques, the digitized images of Alcian blue-stained or 3H-glucosamine-labeled sections digested with an enzyme specific for HA, were subtracted from adjacent, undigested sections. The resultant difference picture images (DPI) accurately depicted the distribution of stained or labeled HA within the cranial mesenchyme. 3H-glucosamine-labeled HA was distributed uniformly throughout the cranial mesenchyme as 12,18, and 24 hr of culture. By contrast, the mesenchyme was uniformly stained with Alcian blue at 12 hr, but stain intensity decreased in the central regions of the mesenchyme

Changes in mesenchymal cell and hyaluronate distribution correlate with in vivo elevation of the mouse mesencephalic neural folds.

The mesenchyme of the elevating mesencephalic neural folds of the mouse is composed primarily of mesenchymal cells embedded in an hyaluronate-rich extracellular matrix. In this study we provide evidence that hyaluronate and mesenchymal expansion may play a role in neural fold elevation and closure. Spatial and temporal patterns of mesenchymal cell and hyaluronate distribution were analyzed duri...

Distribution of Specific Glycoconjugates in Early Mouse Embryonic Notochord and Paraxial Mesenchyme

It is well known that glycoconjugate components of the cell surface and extracellular matrix, play an essential role(s) in many developmental phenomena such as cell differentiation, migration, and cellular interactions. The purpose of this study was to investigate distribution of this macromolecules during differentiation of the notochord and paraxial mesoderm. Formalin fixed paraffin sections ...

Hectd1 regulates intracellular localization and secretion of Hsp90 to control cellular behavior of the cranial mesenchyme

Hectd1 mutant mouse embryos exhibit the neural tube defect exencephaly associated with abnormal cranial mesenchyme. Cellular rearrangements in cranial mesenchyme are essential during neurulation for elevation of the neural folds. Here we investigate the molecular basis of the abnormal behavior of Hectd1 mutant cranial mesenchyme. We demonstrate that Hectd1 is a functional ubiquitin ligase and t...

An Explant Assay for Assessing Cellular Behavior of the Cranial Mesenchyme

The central nervous system is derived from the neural plate that undergoes a series of complex morphogenetic movements resulting in formation of the neural tube in a process known as neurulation. During neurulation, morphogenesis of the mesenchyme that underlies the neural plate is believed to drive neural fold elevation. The cranial mesenchyme is comprised of the paraxial mesoderm and neural c...

CIGARETTE SMOKE/ETHANOL-INDUCED LIMB DEFECTS IN MOUSE EMBRYOS

We have produced a spectrum of limb defects in developing mouse embryos by exposing the mother to smoke from cigarettes with different nicotine concentrations, ethanol, and a combination of ethanol and cigarette smoke. The critical time of exposure was determined to be during day 10 of gestation (vaginal plug=day 0). This time is prior to the critical events which occur between the apical e...
